Kaempferol is a polyphenol belonging to the flavonoid group. The highest concentration of kaempferol (mg/100 g) is found in saffron (12,600), capers (250), arugula or rocket salad (60), kale (48) mustard seed (38), the common bean (26) and endive (25). Organically cultivated plants contain 10% to 95% (e.g., tomatoes) more kaempferol than non-organic plants. Kaempferol, like most flavonoids, is difficult to absorb; its absorption is increased by 52% and 40%, respectively, when combined with natural vitamin E and fat (e.g., coconut oil, olive oil or omega-3 oil). In animals, kaempferol is broken down into quercetin, which is not the case in humans. Kaempferol is readily absorbed through the intestinal wall into the bloodstream by passive transport and then absorbed into the brain. Epidemiological studies show that eating foods rich in kaempferol halves the risk of developing some cancers, cardiovascular diseases and diabetes. Within Chinese medicine, kaempferol is considered a phytonutrient for the treatment of numerous chronic diseases(overview) such as inflammation, cancer, multisystem disorders and degenerative diseases such as diabetes, Alzheimer's, Parkinson's and Huntington's disease. There are more than 5,000 studies published on Pub Med on the action of kaempferol, a small fraction of them by Western scientists. Presumably, kaempferol is relatively unknown in the West for this reason. Epigenetic restoration Kaempferol has the property of restoring the expression of several genes. The balance between turning genes on and off, which makes whether or not certain substances are produced, is regulated by two enzyme groups: the HATand HDAC enzymes. Inhibiting the HDAC enzymes positively affects the expression of genes. HDAC inhibitors increase the expression of the "good genes. There are 12 different HDAC enzymes. Kaempferol has the ability to inhibit 4 HDAC enzymes (HDAC 2, 4, 7 and 8). At the same time, kaempferol increases the action of SIRT3, the enzyme that regulates the production of energy (ATP) and is a central factor in the anti-aging process and the clearance of cancer cells. It also increases the action of the SIRT6 enzyme, which repairs DNA, keeps telomeres intact, regulates sugar burning(glycolysis), clears cancer cells and inhibits inflammation. Properties1. Calming Kaempferol is involved in the regulation of several endogenous substances that calm the nervous system.

  • Glutamate: kaempferol is believed to be the strongest natural glutamate inhibitor (NMDA receptor antagonist) with potency more potent than orally ingested glutamate, GABA, lavender, glycine and magnesium. Glutamate activates psychological and cellular stress and is released as a result of insulin resistance, traumatic experiences, an overloaded stress system and dopamine reward system and upon elaboration of exorphins and comfort foods such as sugar, flavor enhancers, dairy, gluten and soy. Glutamate inhibits dopamine and intensifies negative perception with characteristics such as agitation, brooding, anxiety, irritability, the nocebo effect, learned helplessness, black-and-white thinking and reactive behavior. In people with agitation, brooding, anxiety, stress and sleep disorders, ADHD, depression, eating disorders, burnout and psychological trauma, glutamate is more often chronically elevated.   • GABA: kaempferol is a so-called prodrug, a substance that is converted into active substances via bacteria in the intestines. Kaempferol is converted by bacteria into GABA that is transported to the brain via the vagus nerve in the gastrointestinal system. Studies in animals indicate that a dose starting at 0.02 mg to 1 mg per kg of body weight has a calming effect, which corresponds to 1.5 to 70 mg in a 70 kg person(ref). In people who have taken antibiotics or have disturbed intestinal flora, kaempferol may not be converted into GABA sufficiently or at all (ref).
  • GAD enzyme: this enzyme converts glutamate into GABA, a substance that makes one calm. Kaempferol increases the expression of the GAD65 gene, which regulates the production of the GAD enzyme. Impaired function of this enzyme is responsible, among other things, for diabetes 1 & 2 and the chronic increase in glutamate.
  • Trauma recovery: traumatic experiences and emotional neglect during childhood reduce the action of the GAD enzyme, which is supposed to convert glutamate into GABA. Excessive levels of glutamate cause problems in releasing negative experiences. Research shows that kaempferol has a restorative effect on the GAD65 gene that regulates the production of the GAD enzyme.
  • Sleep process: GABA regulates falling asleep. Deep sleep and the REM phase are regulated by BDNF (see section 4. BDNF).
  • Endorphins: an oral dose of 10 mg kaempferol/kg increases endorphins by 500%.

 

2. Dopamine and serotonin Kaempferol inhibits the enzyme monoamine oxidase A (MAO-A). This inhibits the breakdown of serotonin, norepinephrine and dopamine and increases the availability of these neurotransmitters. The effect is a reduction in depressive symptoms and an improvement in attention and motivation. Because of MAO-A inhibition, kaempferol should not be combined with drugs and dietary supplements with serotonergic effects. Kaempferol also increases dopamine, via the stimulation of anandamide and BDNF. 3. Anandamide Perhaps one of the important functions of kaempferol is to increase anandamide, the bliss substance also released during the "runners high. Kaempferol inhibits the action of the FAAH enzyme, which breaks down anandamide. Research shows that the countries where people are happiest are not the richest countries, but those where people have the highest anandamide levels. The countries where the population has the slowest FAAH enzyme activity - which increases anandamide - are the happiest. According to research , people in Mexico, Nigeria and Colombia are three to six times happier than people in Russia, China and Eastern European countries. In the United States, it is mostly Americans of African and South American origin who have sluggish FAAH enzyme activity; white residents surge at the bottom. Anandamide is an important factor in the perception of spirituality, deep connection, compassion and selflessness. Anandamide increases dopamine and BDNF. 4. BDNF Kaempferol increases BDNF (brain-derived neurotrophic factor), a neurotrophin, which is a neuroprotective substance responsible for the repair and survival of brain cells. A lack of neurotrophins is one of the main causes of neurodegenerative diseases such as Alzheimer's, Parkinson's and Huntington's disease. Without BDNF, some of the brain cells would shrivel and die under the influence of glutamate, something we see, for example, in people with hippocampal atrophy, in which the volume of the hippocampus decreases. BDNF activates the release of GABA and prevents GABA resistance. BDNF has a restorative effect on glutamate, not only because it is a neurotrophic that repairs glutamate damage, but also because it is necessary for GABA function. Without BDNF, GABA would have only a fraction of its action. Thus, to treat glutamate, one must optimize the action of BDNF and GABA simultaneously. The sleep process is regulated, among other things, by GABA and BDNF. GABA is responsible for falling asleep and BDNF for deep sleep and the REM phase. BDNF and dopamine interact: every time dopamine is activated, BDNF is released, which is needed to neutralize the neurotoxicity of dopamine and glutamate. BDNF also enhances the dopamine signal and can restore dopamine sensitivity. Contrary to what the name brain-derived neurotrophic factor suggests, BDNF is active not only in the brain, but also in the lungs. There, it forms a protection against allergic airway diseases. The lungs produce not only BDNF, but also DPP-IV enzymes. These two substances work together to protect the lungs from a variety of immune disorders such as asthma and hay fever. 5. Antioxidant and hydrogen activator Kaempferol stimulates the production of melanin and neuromelanin, the body's strongest antioxidants. Melanin and neuromelanin activate the release of hydrogen, splitting water (H2O) into hydrogen (H2) and oxygen (O) without the help of an enzyme. What does this have to do with reducing oxidative and nitrosative stress? Antioxidants reduce oxidative stress via donating a hydrogen molecule to a free radical. Kaempferol also activates antioxidant enzymes such as superoxide dismutase, catalase and heme oxygenase-1. Kaempferol inhibits pro-oxidant enzymes, which are enzymes that produce oxidative stress such as xanthine oxidase, and eliminates oxidized copper and iron that cause oxidative stress.

Casein, the protein fragment in milk inhibits the antioxidant activity of kaempferol and other polyphenols by up to 75%.

6. Pijnstiller Kaempferol heeft een pijnstillende werking via het herstellen van de expressies van de genen die een pijnremmende functie hebben en het verminderen van pijnmediatoren zoals substance P. Het is geschikt voor het verminderen van chronische en neuropathische pijn, met in het bijzonder migraine, tinnitus, rugpijn en gewrichtspijn. Het pijnstillend effect komt onder meer tot stand door het activeren van anandamide, een van de lichaamseigen cannabinoïden, via een gelijkaardige route die we ook zien bij gebruik van cannabis en PEA. Anandamide activeert tevens de TRPA1-receptoren, die een pijnstillend effect hebben. De pijnstillende werking neemt toe met de tijd door het herstellen van de genexpressies. Neuropathische pijn wordt onder meer veroorzaakt door een combinatie van oxidatieve en nitrosatieve stress, vorming van AGE’s, ontstekingen en toename van glutamaat, een stof die de pijnmediatoren activeert. Kaempferol vermindert deze factoren en is volgens onderzoek een gunstig middel voor het verminderen van neuropatische pijn. 7. Ontstekingsremmend (COX-enzymen) De meest toegepaste werking van kaempferol is het ontstekingsremmende effect. Ontstekingen worden onder meer veroorzaakt door oxidatieve en nitrosatieve stress, ontstekingsbevorderende stofjes, zoals cytokines en het stimuleren van de COX-enzymen, die tevens pijn veroorzaken. Ontstekingsremmende pijnstillers zoals ibuprofen (NSAID), paracetamol en aspirine, werken deels via het remmen van de COX-enzymen. Dit effect wordt deels aangestuurd via het endocannabinoïde systeem, waartoe anandamide behoort. Ontstekingsremmende pijnstillers dragen bij tot endorfine- en anandamideresistentie en een vermindering van glutathion. Volgens het Harm-rapport zijn deze geneesmiddelen verantwoordelijk voor ongeveer de helft van de vermijdbare acute ziekenhuisopnames in Nederland. Het rapport ‘Vervolgonderzoek Medicatieveiligheid’ vermeldt dat het aantal geneesmiddel-gerelateerde ziekenhuisopnamen zijn gestegen van 39.000 in 2008 naar 49.000 in 2013. Kaempferol heeft een ontstekingsremmende werking via het stimuleren van anandamide, een stof die de COX-enzymen remt, wat gunstig is omdat het de vorming tegengaat van prostaglandines, ontstekingsbevorderende stofjes die vaatverwijding, koorts en pijn veroorzaken. Kaempferol remt tevens NF-κB (nucleaire factor kappa B), een stofje dat vrijkomt bij ontstekingen en een rol speelt bij artritis, astma, ziekte van Crohn, multiple sclerose en kanker. Andere ontsteking-gerelateerde stoffen die door kaempferol worden geremd zijn TNFα en Interleukine-6 (IL-6). Uit onderzoek is aangetoond dat kaempferol nuttig is voor de behandeling van onder meer: chronische en laaggradige ontstekingen, maag- en darmontstekingen, colitis ulcerosa, IBS, artrose, (reumatische) artritis, borstontsteking, maagzweren, ontsteking van de bloedvaten en acute aandoeningen zoals ARDS en longontsteking. 8. Insuline en glucose Kaempferol verbetert de werking van insuline en de glucoseverbranding via meerdere routes. Het bevordert de werking van insuline, onder meer door toename van IRS-1 (insulin receptor substrate-1 ) een eiwit dat de insuline signaaloverdracht doorgeeft in de cellen. Laaggradige ontsteking zijn een belangrijke oorzaak (en gevolg) van pre-diabetes, ofwel vergevorderde insulineresistentie. Twee factoren spelen hierbij een belangrijke rol: IκB kinase, een stof die het insulinesignaal blokkeert en NF-κB stimuleert, dat verschillende ontstekingsstofjes activeert, beide factoren worden door kaempferol afgeremd. Kaempferol is een insuline-imitator, met onder meer een verhoogde opname van glucose tot gevolg. Kaempferol verbetert de glucosetolerantie, dit is de mate dat het lichaam glucose kan verwerken, door het verhogen van de insulineaanmaak, een verbeterde glucoseopname, een verminderde vrijgave van glucose via de lever (gluconeogenese) en het verlagen van de bloedsuikerwaarden. Kaempferol vermindert stikstofmonoxide (NO, nitric oxide) en oxidatieve stress, twee belangrijke oorzaken van insulineresistentie. Het vermindert tevens de symptomen van het metabool syndroom zoals toename van HDLen vermindering van LDL. 9. Geheugen en hippocampus Kaempferol verbetert het geheugen, onder meer door de cellen te herstellen in de hippocampus, die beschadigd zijn t.g.v. glutamaat gerelateerde hippocampusatrofie. 10. Antidepressant (BDNF en anandamide) Kaempferol heeft een antidepressief effect via de toename van anandamide en BDNF (zie punt 3 en 4). 11. Antihistaminicum Kaempferol remt de vrijgave van histamine uit de mestcellen en heeft een gunstig effect bij de behandeling van IgE-gemedieerde aandoeningen zoals astma, hooikoorts en andere allergische luchtwegaandoeningen. 12. Botdensiteit (calcium, osteoporose) Menopauzaal botverlies ontstaat door afname van oestrogeen en progesteron. Onderzoek toont aan dat kaempferol de oestrogeen en progesteron huishouding helpt te herstellen en gunstig is om de botdensiteit in stand te houden. Bij botontkalking door bijvoorbeeld hypercortisol, medicatie, metabool syndroom, alcoholisme, roken en diabetes blijkt kaempferol een gunstig effect te hebben op de botdensiteit. Onder meer door het verminderen van osteoclasten (cellen die het bot afbreken) en toename van osteoblasten (cellen die het bot opbouwen). Onderzoek wijst uit dat kaempferol een gunstig effect heeft in het herstelproces van ruggenwervel-degeneratie, waarbij de tussenwervelschijven zijn aangetast, bijvoorbeeld door botontkalking of fysieke belasting. Kaempferol heeft een gunstig effect in het versnellen van het genezingsproces bij botbreuken, de aangroei van implantaten en het herstel van het kraakbeen via het stimuleren van de chondroblasten en chondrocyten. 13. Kankerwerende eigenschappen Uit epidemiologische studies blijkt dat het eten van kaempferolrijke voeding het risico op het ontwikkelen van sommige kankers, cardiovasculaire aandoeningen en diabetes halveert. Kanker wordt onder meer veroorzaakt door defecten in het reproduceren van DNA, die op hun beurt ontstaan door oxidatieve en nitrosatieve stress. Kaempferol sti­muleert de aanmaak van drie lichaamseigen stofjes die de DNA-fouten eruit filteren: anandamide, BDNF en endorfine. Kaempferol is een chemopreventieve stof die ingrijpt in de verschillende fasen in de ontwikkeling van kanker. Het preventief anti-kankereffect is onder meer aangetoond bij slokdarmkanker, borstkanker, baarmoederhalskanker, leverkanker, eierstokkanker, maagkanker, longkanker, leukemie, galwegkanker, alvleesklierkanker, blaaskanker, botkanker en goedaardige vleesbomen in de baarmoeder (uterus myomatosus). Kaempferol vermindert HTLV-1, dit is een oncovirus, een virus dat kanker veroorzaakt zoals leukemie. 14. Hart en bloedvaten Kaempferol voorkomt en helpt atherosclerose te herstellen, deels door de oorzaak (geoxideerde LDL cholesterol) aan te pakken. Kaempferol voorkomt de vorming van bloedklonters (trombose) en plakvorming in de bloedvaten. Kaempferol heeft een beschermende functie tegen het ontstaan van aritmieën, ventrikelhypertrofie en hartinfarct. 15. Antibacterieel Kaempferol heeft een antibacteriële werking, onder meer tegen Helicobacter pylori die bij sommige mensen kan leiden tot maagzweren, ontstekingen van het maagslijmvlies en twaalfvingerige darm en maagkanker. Het biedt tevens bescherming tegen Escherichia coli, Pseudomonas aeruginosa, Proteus mirabilis en Staphylococcus aureus. 16. Antiviraal, antischimmel en eencellige parasieten Kaempferol heeft in onderzoek aangetoond de replicatie van sommige virussen (het kopiëren van het virale DNA in de cellen) te stoppen of af te remmen, voornamelijk via het remmen van enzymen die de replicatie activeren zoals transcriptase, viral proteases en neuraminidase. Het remmend effect is aangetoond bij het herpes simplex virus, cytomegalovirus, influenza virus (griep) en HIV (Aids). Kaempferol helpt bij het neutraliseren van enzymen die door de Candida albicans wordt afgescheiden om de cellen van de gastheer te penetreren, om op deze manier de opname van C. albicans in de cellen te voorkomen. Uit onderzoek blijkt dat kaempferol gunstig is bij de behandeling van de schimmels Aspergillus flavusAspergillus niger en Trichophytonmentagrophytes. Kaempferol heeft een antiprotozoale functie, dit is het vernietigen van eencellige parasieten zoals Leismania sppEntamoeba histolytica en Giardia lamblia. 17. Vetcellen Kaempferol remt FASN (Fatty Acid Synthase), een enzymgroep die glucose omzet in palmitinezuur, een verzadigd vetzuur dat in te hoge concentraties in verband wordt gebracht met overgewicht, moeite met afvallen en kanker. Palmitinezuur dat het belangrijkste vetzuur van palmolie is, veroorzaakt nadelige epigenetische aanpassingen in de cellen. Stamcellen die in een later stadium spiercellen of vetcellen worden, de zogenoemde ‘precursor-cellen’, veranderen in een omgeving met veel palmitinezuur in vetcellen. Kaempferol draait het proces om, zodat de precursor-cellen veranderen in spiercellen. 18. Vermindering van AGE’s Kaempferol zorgt voor een aanzienlijke afname van AGE’s, afkorting voor Advanced Glycation End-products, zijn eiwitten en vetten die onherstelbaar beschadigd zijn, doordat ze op een verkeerde manier zijn gekoppeld aan suikergroepen zoals glucose. Hierdoor zijn de gedegenereerde eiwitten en vetten niet meer bruikbaar en kunnen ze een gevaar vormen. AGE’s worden deels verantwoordelijk geacht voor het ontstaan van microvasculaire complicaties bij hyperglycemische aandoeningen als diabetes. AGE’s veroorzaken een toename van oxidatieve stress, insulineresistentie en chronische ontstekingen. Ze veroorzaken een versnelde verharding van de pezen in de spieren, waardoor schouders en handen hun soepelheid verliezen. Referenties
